TGF-β1是一组蛋白超家族。TGF-β1是由两个结构相同或相似,分子量为12.5kD的亚单位通过二硫键连接而成的双体。在人的心肌成纤维细胞、血管内皮细胞和巨噬细胞等均有表达。TGF-β1信号通路的关键传导分子为胞质蛋白Smads,他们可以将TGF-β1信号直接由细胞膜受体传导人细胞核内,是受体激酶介导的细胞内信号传导的新途径,为细胞信号跨膜后与核内基因转录之间开通了一条简便路径。不同的Smad介导不同的TGF-β1家族成员的信号转导。TGF-β1/Smad信号转导通路活化后,可以促进相关基因的转录,如胶原蛋白的表达增高,也可通过调节MMPs/TIMPs表达的平衡抑制胶原降解,促进组织纤维化。订购咨询电话:18580872564  QQ:481279580

Members of TGF-family are multifunctionalcytokines that regulate cell proliferation, growth, differentiation andmigration as well as synthesis of the extracellular matrix. The three humanisoforms, TGF- 1,  2,  3, signal through the same receptor andelicit similar biological responses. The release of biologically active TGF-isoformfrom a latent complex involves proteolytic processing of the complex. TGF-1 isthe most abundant isoform secreted by almost all cell types. Human TGF-1 is a25.0kDa protein with each subunit containing 112 amino acid residues, linked bya disulfide bond.

• Biological Activity: The activity was determined by using a SMADreporter gene assay in human 293 cells. The EC50 ranges from 0.1 – 1.0 ng/ml.

ne of the three closely-related members of the TGF-β family. Human TGF-β1 cDNA encodes a 390 aa precursor with a 29 aa signal peptide. A furin-like convertase cleaves the precursor to form a 249 aa latency-associated peptide (LAP) and a C-terminal 112 aa mature TGF-β1. Disulfide-linked mature TGF-beta1 is dissociated from the LAP upon activation.
StemRD, produces active TGF-β1 in human 293 cells in Xeno-free medium. After a series of chromatography, the protein is purified to near homogeneity. Activity is determined by using a SMAD reporter gene assay with EC50 0.1 – 1.0 ng/mL.
